The success of a private review class for candidates appearing for the pharmacy board at Liceo de Manila in 1903 led to a surge of interest for such lectures among the candidates and a few more wanted to join. The number of students enrolling suddenly increased and Escuela de Farmacia (The founder) had to ask all his graduates to come over and educate the new group of candidates, which further led to the establishment of Manila College of Pharmacy in 1915. The Dentistry College started in 1929 and then it was renamed as Manila College of Pharmacy & Dentistry. During the war the University was closed but then later started again in 1945.

The University has received accreditation from the Commission on Higher Education, Philippines as well as the Philippine Association of Colleges and Universities Commission on Accreditation (PACUCOA). The MCU College of Nursing was ranked as the 4th Top Performing Nursing School in the Philippines by the Professional Ranking Commission (PRC) in the year 2009 and the College of Optometry was positioned as the country’s Top Performing Optometry School by the same body in the year 2013-14.
